What is your character's relation to and opinion of every other character they interact with and talk about? Relationships are fundamental.
The script is an actor's bible. Every word counts. Make sure you read and absorb all of the information on all pages for full clarity.

Don't let the crew or gear distract you for even a moment. The only people you think about are actors in your scene.
Motives. What motivates your character's actions? A little psychological character development and history mapping can go a long way.
Some people choose to be actors and others choose to be dramatic. Choose wisely.

Arriving early to a set or audition is a great way to get settled before you work. It helps maintain focus & allows time for any mistakes.
It's ALWAYS about the performance, but it doesn't hurt to know who you're performing to.
If it's a drama and the writer is in the room, you should probably be much closer to the exact dialogue.
Generally speaking, if it's a comedy and a first audition, you can play with the words & delivery quite a bit (& should, to help stand out).
