Cancer researchers, what's your best take on the definition of cancer? I've been pondering this question and I’d love to hear from others
.@EmilyAshkin et al @LabWinslow underscore important roles of STAG2 in suppressing lung tumorigenesis & highlight a STAG2-PAXIP1/PAGR1 tumor-suppressive program that may transcend cancer type. hubs.la/Q02-Y7_D0 @HaiqingXu3 @kinglhung @HowardYChang @Jesse_R_Dixon @ZXuSalk
#PCAMChat24 A3: From a clinical/translational standpoint, we finally have drugs that can target KRAS in #PancreaticCancer. This gene is abnormal in >90% of cases, and it has taken four decades since the original discovery tofinally have a slew of drugs in the pipeline that can…
“There are not, and will never be, KRAS inhibitors” 👀 @Pasca_Lab presents a memorable reviewer comment from her 1st unfunded NIH grant at #AACRPan24. The lesson is that you should not let deflating reviewer comments define the value of your science. Also, persistence!
The company ran a 240 patient (!) randomized phase 2 trial of Simtuzumab in #PancreaticCancer that showed no therapeutic benefit. academic.oup.com/oncolo/article… As an angiogenesis inhibitor (versus the stated inhibition of LOXL2 catalytic activity) this was essentially a rerun of Gem…

Agreed. I’ve always thought that cancer is when cellular fitness becomes dominant over organismal fitness.
For an overarching theme I propose "Reneging on a contract that balances cellular and organismal fitness."

I consider it a disease of progressive tissue dysregulation that leads to eventual systemic failure. But I draw heavily from @TuftsUniversity Drs. Soto and Sonnenschein “TOFT” theory which IMO is by far the best explanation I’ve seen yet: ncbi.nlm.nih.gov/pmc/articles/P…
I usually define it as cell selfishness, in which individual cells evolve to become independent from the rest, but taking advantage of the shared resources.
A mutation+evolution driven state of enhanced fitness/robustness relative to its normal counterpart (at the cellular or tissue level). This happening at such an incredibly accelerated and uncoordinated manner at the expense of the host.
A reversion from multicellular, diploid, sexual reproduction to unicellular, diploid, asexual reproduction. Metastasis, dormancy, and other cancer behaviors are best explained by evoluationary theory applied to individual cells residing the environment of the body.
At its core, a disorder characterized by cell uncontrolled proliferation
