Great news, this does not only work in fixed cells but also in ultrathin cryosections of tissues, as we show for tokPAINT imaging of Pol II within mouse tissues. [5/11]

Achieving sub-3 nm localization precisions, tokPAINT enables detection of Pol II down to level of single antibodies. Bonus: cryosections are openly accessible to probe diffusion, enhancing imager binding kinetics, e.g. for molecular counting via qPAINT. [4/11]
Inspired by previous work (e.g., by @apombo1, @Eric_Betzig, @HeilemannLab) we adopted Tokuyasu cryosectioning, widely known from Immunogold EM, to enable nuclear tokPAINT imaging of Pol II using TIRF. [3/11]
